Semaglutide is a once weekly GLP-1 agonist. Enter your vial, the bacteriostatic water you add and your weekly dose to see exactly how many units to draw.
= 0.25 mg | Weekly dose. Most protocols start at 0.25 mg (250 mcg).
1 mg = 1,000 mcg (micrograms). Most peptide protocols are written in mcg.
Common vial sizes are 3, 5 and 10 mg.
For a 10 mg vial mixed with 2 mL bacteriostatic water (5.00 mg/mL), on a U-100 insulin syringe.
| Dose | Volume | Units | Doses per vial |
|---|---|---|---|
| 250 mcg | 0.050 mL | 5u | 40 |
| 500 mcg | 0.100 mL | 10u | 20 |
| 1 mg | 0.200 mL | 20u | 10 |
| 1.7 mg | 0.340 mL | 34u | 5 |
| 2.4 mg | 0.480 mL | 48u | 4 |
Units shown for your current vial and water amount (10 mg / 2 mL).
| Weeks | Weekly dose | Units to draw |
|---|---|---|
| 1 to 4 | 250 mcg | 5u |
| 5 to 8 | 500 mcg | 10u |
| 9 to 12 | 1 mg | 20u |
| 13 to 16 | 1.7 mg | 34u |
| 17+ | 2.4 mg | 48u |
Based on clinical titration. Stay at a step longer if side effects are strong.
Semaglutide doses are small, so the right water amount matters. With a 10 mg vial and 2 mL of water, a 0.25 mg starting dose is only 5 units, so many people add more water to make small doses easier to measure.
Titration steps up every 4 weeks: 0.25, 0.5, 1, 1.7 and 2.4 mg. Inject once a week on the same day and store the mixed vial in the fridge.
Every step below uses the numbers you entered above.
Swab the rubber tops of the 10 mg Semaglutide vial and the bacteriostatic water vial with fresh alcohol pads. Let them air dry for a few seconds.
Pull 2 mL of bacteriostatic water into a reconstitution syringe (usually a 3 mL syringe with a 21g needle).
Inject the water against the inside wall of the vial, don't blast it directly onto the powder. This protects the peptide from shear damage.
Gently swirl until the powder is fully dissolved and the liquid is clear. Never shake, foaming denatures the peptide.
On your 100-unit (1 mL) insulin syringe, pull the plunger back until the rubber tip lines up with 5, that's your 250 mcg dose. Tap out any air bubbles, then inject as directed.
Cap the vial and store at 2–8 °C (36–46 °F). Most reconstituted peptides remain stable for 3–4 weeks refrigerated.
With a 10 mg vial and 2 mL of water (5 mg/mL), 0.25 mg is 0.05 mL, or 5 units. With a 5 mg vial and 2 mL, it is 10 units.
2 mL is common and gives 2.5 mg/mL, so 0.25 mg is 10 units and 1 mg is 40 units.
0.25 mg weekly for 4 weeks, then 0.5 mg, 1 mg, 1.7 mg and 2.4 mg, increasing every 4 weeks as tolerated.
Semaglutide targets GLP-1 only. Tirzepatide targets both GLP-1 and GIP and showed greater average weight loss in head to head trials.
